eSATA on mobo?

I'm not sure what you mean.

 

If you mean the E-Sata port on the case, it will need to be connected to one of the sata ports on your motherboard (If you want it to work).

 

If you mean the E-Sata port on the motherboard, it does not need to be connected to any sata ports.
